Enter at street level at 356 W 58th St, take the escalator up to the lobby. From facing the lobby, turn left to see the elevators, walk towards them, then instead of getting in them, turn right down a hallway and take your first left, there are two genderfree restrooms.
There is a bar behind the check-in desk (where you walk in), so coming in late-ish at night is probably fine! The lobby is pretty cool looking, and the bathrooms are huge for NYC standards.
